Abstract (eng): | Drug delivery matrices are three-dimensional structures consisting of polymers, dendrimers or surfactants. Nanoscale matrices have the potential to solve problems related to drug delivery and targeting especially in cancer therapy. Many cancer drugs are toxic for healthy tissues and have strong side effects. Encapsulation matrices enable drug targeting to the diseased cells which reduces the side effects and the amount of drug needed as well as improves the therapeutic effect. There are two targeting methods for the matrices, active and passive targeting. Active targeting can be achieved with certain ligands and antibodies whose receptors are overexpressed in cancer cells. Passive targeting is based on the anatomical differences between normal and cancer tissues. Drug release from the matrix can be controlled with different physical properties such as temperature and ph. Additionally certain imaging agents can be attached to the matrices to monitor the therapy. Liposomes, polymeric micelles and dendrimers are common nanoparticles used in drug delivery. The small size of the nanoparticles is their advantage and enables them to reach even the smallest junctions of the body. The drug can be encapsulated or covalently bounded to the nanoparticle. Some pharmacokinetic characteristics that the matrices should possess are prolonged circulation, stability in blood circulation and biocompatibility. 5-Fluorouracil is a common cancer drug, which is due to its toxicity almost only used as a prod rug. In the experimental part two syntheses of two 5-fluorouracil derivatives are presented. Both derivatives have potential to function in a cancer drug application based on a drug delivery matrix. |
Abstract (fin): | Lääkekuljetukseen tarkoitetut kapselointimatriisit ovat kolmiulotteisia rakenteita, jotka koostuvat polymeereistä, dendrimeereistä tai surfaktanteista. Nanokokoluokan matriisit ovat yleistyneet viime vuosina ja varsinkin syövän hoitoon tarkoitettuja matriiseja tutkitaan paljon. Ongelmana monien syöpälääkkeiden käytössä on niiden toksisuus terveille kudoksille ja vahvat sivuvaikutukset. Matriiseilla pystytään kohdentamaan lääkeaine tiettyyn kohtaan elimistössä, jolloin sivuvaikutukset vähenevät, vaikutus tehostuu ja lääkeainetta tarvitaan vähemmän. Matriiseilla tapahtuva kohdennus jaetaan aktiiviseen ja passiiviseen kohdennukseen. Aktiivinen kohdennus voidaan saavuttaa ligandien ja vasta-aineiden avulla, joiden reseptoreja esiintyy paljon syöpäsolujen pinnalla. Passiivinen kohdennus perustuu normaalin ja syöpäkudoksen anatomisiin eroihin. Lääkeaineiden vapautumista matriisista voidaan kontrolloida erilaisten fysikaalisten ominaisuuksien avulla, kuten lämpötila ja pH. Lisäksi matriiseihin liitetään usein kuvantamisyksikoitä, joilla kohdentumista voidaan seurata. Liposomit, polymeeriset misellit ja dendrimeerit ovat yleisiä lääkeainekuljetuksessa käytettyjä nanopartikkeleita. Nanopartikkelien etuna on niiden pieni koko, jonka ansiosta ne pääsevät kulkeutumaan elimistön ahtaisiin kohteisiin. Lääkeaine voidaan joko kapseloida tai liittää kovalenttisesti nanopartikkeliin. Farmakokineettisiä vaatimuksia lääkekuljetusmatriiseille ovat muun muassa pitkitetty kierto, stabiilisuus verenkierrossa ja bioyhteensopivuus. 5-Fluorourasiili on yleinen syöpälääke, jota toksisuutensa ansiosta käytetään lähes ainoastaan aihiolääkkeenä. Kokeellisessa osassa kuvataan kahden erilaisen 5-fluorourasiilijohdannaisen synteesi, joilla on potentiaalia toimia lääkekuljetusmatriisiin pohjautuvassa syöpälääkkeessä. |
