Abstract (eng): | In this thesis, nine microbiological modelling programs were investigated. The aim of the study was to go through different modeling programs and to choose the most appropriate one regarding Valio Ltd.'s requirements. The suitability and functionality of the modeling program was demonstrated by practical studies. Both the effect of thermal treatment on Escherichia coli bacteria and the determination of the growth and growth rate of Salmonella Abony bacteria were investigated. Microbiological modelling programs simulate responses of micro-organisms in different environments. Modelling programs are based on mathematical models which are used to predict the growth or destruction of pathogenic or spoiler micro-organisms. Sym'Previus and Com Base programs were found to be the most appropriate ones for Valio Ltd because they demonstrate the growth and heat destruction of microorganisms. The suitability of these programs to predict the effect of thermal treatment of microorganisms was investigated by a heater that heated the milk and cream samples contaminated by Escherichia coli bacteria. The suitability of modeling programs to predict the growth of microorganisms was investigated by buffered peptone water samples contaminated by S. Abony bacteria. The results of the experimental work were set against the predictions of modeling programs in the corresponding environment circumstances. The results of thermal treatments did not meet the predictions of modeling programs. The difference may be due to the heater or the E. coli strain used in this work that may differ the E. coli strain used in modeling programs. Com Base program was found to predict well the growth of Salmonella bacteria but Sym 'Previus program did not predict that properly in our case. It would be advisable to use several modeling programs to predict the growth and destruction of microorganisms because the strains of bacteria differ from each other and the predictions of modeling programs are not completely reliable. |
Abstract (fin): | Tässä työssä käytiin läpi yhdeksän saatavilla olevaa mikrobiologista mallinnusohjelmaa. Työn tarkoituksena oli etsiä parhaiten Valio Oy:n käyttöön soveltuva mallinnusohjelma ja todentaa sen toimivuus käytännön koejärjestelyillä. Mikrobiologisilla mallinnusohjelmilla simuloidaan mikrobien vasteita eri ympäristöolosuhteissa. Mallinnusohjelmat perustuvat matemaattisiin malleihin, joiden avulla voidaan ennustaa esimerkiksi patogeenisten tai pilaajamikrobien kasvua ja tuhoutumista eri olosuhteissa. Parhaiten Valio Oy:n käyttöön todettiin soveltuvan Sym'Previus- ja ComBase-ohjelmat, sillä ne mallintavat sekä patogeenisten että pilaajamikrobien kasvua ja tuhoutumista. Mallinnusohjelmien soveltumista lämpökäsittelyn vaikutuksen ennustamiseen tutkittiin kuumennuskäsittelylaitteella, jolla kuumennettiin keinotekoisesti Escherichia coli -bakteereilla kontaminoituja maito- ja kermanäytteitä. Mallinnusohjelmien soveltumista mikrobien kasvun ennustamiseen tutkittiin Salmonella Abony -bakteereilla keinotekoisesti kontaminoiduissa puskuroiduissa peptonivesissä. Kokeellisten osien tuloksia verrattiin mallinnusohjelmien ennusteisiin vastaavissa ympäristöolosuhteissa. Kokeellisissa kuumennuskäsittelyissä E. coli -bakteereista tuhoutui huomattavasti suurempi osa mallinnusohjelmien ennusteisiin verrattuna. Ero saattaa johtua esimerkiksi käytetystä kuumennuskäsittelylaitteesta tai E. coli -kannasta, joka luultavasti erosi mallinnusohjelmien käyttämistä E. coli -kannoista. ComBase-ohjelman todettiin ennustavan hyvin Salmonella-bakteerien kasvua, mutta Sym'Previus-ohjelman ei todettu soveltuvan hyvin tässä työssä tehtyyn kasvun simulointiin, sillä simulointia varten oli tiedettävä mikrobin optimaalinen kasvunopeus ja minimi lag-aika tutkitussa näytemateriaalissa. Koska mallinnusohjelmat perustuvat matemaattisiin malleihin ja niissä käytetyt mikrobikannat eroavat toisistaan, on hyvä käyttää rinnakkain useampaa mallinnusohjelmaa ennustamaan mikrobien kasvua ja lämpökäsittelyn aikaansaamaa tuhoutumista. |
