Abstract (eng): | The aim of this study was to investigate the removal of residual fat and mould spores from cheese whey by microfiltration. The primary objective was to find a suitable pore size ceramic microfiltration membrane to maximize the amount of the residual fat removed without a significant protein loss. The second objective of this study was to remove mould spores from whey. Yearly about 800 000 tons of whey from Valio Ltd.'s cheese production is used to whey powder production. The theoretical potential for recovery of fat from already separated whey is about 400 tons. In addition, Aura-cheese (blue cheese) whey is currently used as animal feed because of its supposed spore content. If the spores were removed, about 9 000 tons of Aura cheese whey could be used to produce 500 tons of demineralized whey powder and 11 tons of whey fat. In addition to the economic importance, there is also an industrial importance: Whey fat poses challenges to powder production processes and may cause flavour changes to the end-product when oxidizing. The study was carried out by using a pilot-scale microfiltration equipment with multiple ceramic microfiltration membranes of different pore sizes. Used pore sizes were 0.1, 0.5, 0.8, 1.4, 2.0 and 5.0 µm. A number of different pore size membranes were used to get comprehensive information of protein and fat permeation. In addition, the parallel filtering tests were carried out to ensure the reproducibility of the test runs. To avoid challenges in the transport and storage of the raw materials, reconstituted whey powder was used in the test runs. However, the most significant results were confirmed by using fresh whey. The obtained filtration fractions were analysed for fat, protein and dry matter contents and these results were used to determine the yields of fat and protein to permeate and retentate fractions. On the basis of the results of this study it is possible to remove fat completely only by using a 0.1 µm pore size membrane. However, the protein loss with above was over 55 %. For technical reasons, the minimum protein loss was 10 % which was achieved by using the membranes with pore sizes of 1.4, 2.0 and 5.0 µm. Nevertheless, with those membranes the defatting efficiency was at its best only about 35 %. According to the results, 0.8 µm pore size membrane would be the most suitable for the defatting of whey because its defatting efficiency was about 50 % and protein loss was only 25 %. In addition, it was found out that the membranes with pore sizes of 0.5, 0.8 and 1.4 µm retained mould spores completely. Based on the research, the separation of fat from whey, at least partially, is possible without significant loss of protein. It was also found that the removal of mould spores can also be performed by microfiltration. As a conclusion, both of these research objectives were achieved, but further investigation is required. |
Abstract (fin): | Tämän työn tarkoituksena oli tutkia rasvan ja homeitiöiden erottamista juustoherasta mikrosuodatuksen avulla. Työn päätavoitteena oli huokoskooltaan sopivan keraamisen mikrosuodatuskalvon valinta siten, että mahdollisimman suuri osa separoidun heran jäännösrasvasta saadaan erotettua niin, ettei merkittävää proteiinihävikkiä aiheudu. Toisena tavoitteena oli homeitiöiden erottaminen herasta. Valio Oy:n juustonvalmistuksessa syntyy vuosittain heraa noin 800 000 tonnia. Separoinnin jälkeen heran rasvapitoisuus on noin 0,05 %, jolloin separoidun heran teoreettinen rasvan talteenottopotentiaali on noin 400 tonnia. Lisäksi nykyisin rehukäyttöön toimitettavaa Aura-juustoheraa syntyy vuosittain noin 9 000 tonnia, josta voitaisiin homeitiöt poistamalla valmistaa 500 tonnia demineralisoitua herajauhetta sekä erottaa 11 tonnia rasvaa. Taloudellisen merkityksen lisäksi rasvanpoistolla on teollista merkitystä, sillä heran rasvat aiheuttavat haasteita jauheenvalmistusprosesseissa. Tutkimus suoritettiin suodattamalla heraa pilot-mittakaavan mikrosuodatuslaitteistolla käyttäen useita eri huokoskoon keraamisia mikrosuodatuskalvoja. Käytetyt huokoskoot olivat 0,1, 0,5, 0,8, 1,4, 2,0 ja 5,0 µm. Usean eri huokoskoon suodatuskalvon käytöllä haluttiin varmistaa kattavan tulosaineiston muodostuminen. Lisäksi rinnakkaisilla suodatuskoeajoilla varmistettiin koeajojen toistettavuus. Raaka-aineina koeajoissa käytettiin kuljetus- ja säilytyshaasteiden välttämiseksi herajauheesta ennastettua heraa. Merkittävimmät tulokset kuitenkin varmistettiin käyttämällä tuoretta heraa. Suodatuskoeajoissa saaduista jakeista analysoitiin rasva-, proteiini- ja kuiva-ainepitoisuudet, joiden perusteella määritettiin rasvan ja proteiinin jakautuminen kalvon läpäisseeseen jakeeseen eli permeaattiin ja läpäisemättömään jakeeseen eli retentaattiin. Saatujen tulosten perusteella vain huokoskoon 0,1 µm suodatuskalvo pidätti rasvan täydellisesti. Tuolloin proteiinihävikki oli kuitenkin jopa yli 55 %. Laiteteknisistä syistä pienin saavutettavissa oleva proteiinihävikki oli 10 %, johon päästiin huokoskokojen 1,4, 2,0 ja 5,0 suodatuskalvoilla. Rasvanpoistotehokkuus oli tuolloin kuitenkin enintään 35 %. Yhteisominaisuuksiltaan parhaaksi suodatuskalvoksi todettiinkin siten huokoskoon 0,8 µm kalvo, jolla proteiinihävikki oli 25 %, mutta rasvanpoistotehokkuus 50 %. Lisäksi itiöidenpoistokokeiden perusteella todettiin, että kaikilla testattujen huokoskokojen suodatuskalvoilla homeitiöt pidättyivät täysin. Tutkimuksen perusteella rasvan erottaminen herasta ainakin osittain on mahdollista ilman merkittävää proteiinihävikkiä. Lisäksi todettiin, että homeitiöiden poistaminen voidaan myös suorittaa mikrosuodatuksen avulla. Homepitoisuusanalyyseistä tosin selvisi, että homeitiöt pidättyvät jo heran separointivaiheessa, joten mikrosuodatuksen suorittaminen pelkästään itiöiden poistamiseksi ei ole tarpeellista. Molemmat tutkimustavoitteet saavutettiin, mutta lisätutkimuksia tarvitaan menetelmän teollista hyödyntämistä varten. |
