Abstract (eng): | The so called lecithin organogels are viscous solutions of organic solvent and polymer-like self-assembled structures of phospholipids. The gelation process is controlled by a polar substance, most commonly water. These systems have promising applications in templating nanomaterials and as a basis of functional materials capable of reacting into changes in environmental factors. However, before the full potential of these systems can be utilized, a more clear picture of the gelation process than experimental methods can provide is needed. Recently computational molecular modeling has become popular especially in studying research topics where experimentation is difficult or impossible. On the other hand modern molecular modeling methods suitable for simulating phospholipid systems are typically parameterized to reproduce the behaviour of lipids in aquaeous bilayer structures and it is yet unclear how well these methods perform in organic media. The purpose of this thesis is to investigate how well a molecular model parameterized in aquaeous environment performs in reproducing the behaviour of organic reverse micellar systems. Furthermore, the dynamics and structure of water inside phospholipid reverse micelles was characterized under three hydration levels and the effect of salts NaCl and CaCl2 on the shape of the reverse micelles was investigated. It was found that CHARMM-lipid model has too high tendency towards bilayer structure in organic media and that calcium ions overbind to the lipids under low hydration conditions. Furthermore, it was found that lipid hydration in reverse micelles closely resembles bilayer hydration. Hydrogen bonds lifetimes and interlipid hydrogen bonding decrease as a function of head group hydration. Based on the results the organogel formation in cyclohexane was deemed to be a result of the apparent shape of the lipids changing due to hydration layer build up. |
Abstract (fin): | Niin kutsutut lesitiiniorganogeelit ovat viskooseja liuoksia, jotka koostuvat orgaanisesta liuottimesta ja polymeerimäisistä, itsejärjestäytyneistä fosfolipidirakenteista. Gelaatioprosessia hallitsee poolinen yhdiste, yleisimmin vesi. Organogeelit ovat lupaavia systeemejä huokoisten ja pitkulaisten nanomateriaalien valmistukseen sekä erilaisten herätteisiin reagoivien materiaalien pohjaksi. Ennen kuin organogeelejä voidaan kunnolla hyödyntää, tarvitaan kokeellisten menetelmien antamaa tarkempi kuva gelaatioprosessista. Hiljattain laskennallisista molekyylimallinnusmenetelmistä on muodostunut tärkeä työkalu erityisesti kokeellisesti vaikeasti mitattavien kohteiden tutkimiseen. Toisaalta lipidisysteemien mallintamiseen soveltuvat laskennalliset mallit on tyypillisesti parametrisoitu toistamaan molekyylien käytös vesiympäristössä ja on epäselvää miten hyvin mallit toimivat orgaanisessa liuottimessa. Tämän diplomityön tarkoituksena on tutkia miten hyvin vesiympäristöön parametrisoitu lipidimalli onnistuu toistamaan molekyylien käytöksen organogeelin kaltaisessa käänteismisellisysteemissä. Lisäksi käänteismisellien sisällä olevan veden rakenne ja dynamiikka karakterisoidaan kolmella hydraatioasteella sekä suolojen NaCl ja CaCl2 vaikutus käänteismisellien muotoon selvitetään. Tuloksista havaitiin, että CHARMM-lipidimallilla on liiallinen taipumus kaksoiskerrosmaiseen rakenteeseen orgaanisessa liuottimessa ja lisäksi kalsiumionien havaittiin ylisitoutuvan lipidien pääryhmiin matalalla hydraatioasteella. Lipidien hydraatio käänteismiselleissä muistuttaa läheisesti lipidien hydraatiota kaksoiskerroksissa. Vesimolekyylien dynamiikkka hidastuu sekä lipidien välisten vetysidossiltojen määrä pienentyy hydraation funktiona. Tulokset viittaavat siihen, että sykloheksaani-liuotinsysteemissä organogeelin muodostuminen johtuu lipidien pääryhmän ympärille kasvavan hydraatio kerroksen lipidin näennäistä muotoa muuttavasta vaikutuksesta. |
