Abstract (eng): | Leishmaniasis is a tropical disease caused by parasites of the genus Leishmania. It is transmitted by sand-flies. Leishmaniasis affects the poorest of the poor mainly in the third-world countries. Four main types of leishmaniasis exist and one of them, visceral leishmaniasis, is fatal if left untreated. Visceral leishmaniasis is caused by L. donovani in East Africa and the Indian subcontinent, and by L. infantum in the Mediterranean region and Latin America. The life cycle of a Leishmania parasite consists of two forms: a flagellar promastigote form that lives in the sand-fly midgut and an amastigote form that lives and proliferates in mammalian white blood cells. All commercial antileishmanial medications have severe side-effects and need to be administered under supervision of medical personnel. The growing occurrence of resistant parasites is ever increasing the demand for new and effective antileishmanial medications. The aim of this study was to synthesize a series of benzoxadiazole derivatives, because compounds of similar structure had earlier been noted to possess antileishmanial activity against L. donovani. These derivatives were then to be tested in vitro against L. donovani axenic amastigotes by a cooperation partner. Ten benzoxadiazole derivatives were successfully synthesized in this thesis. Some of the derivatives were synthesized according to the previously designed seven-step synthetic route, but for some a new synthetic route was o designed. All synthetic routes are described in this thesis. Unfortunately, the antileishmanial activity results were not yet available by the time this thesis was written so the structure-activity relationships could not be analysed. However, the information these compound may give about the antileishmanial activity of compounds with similar structure is discussed. |
Abstract (fin): | Leishmaniaasi on Leishmania-sukuun kuuluvien alkueläinten aiheuttama trooppinen tauti, joka leviää hietakärpäsen välityksellä. Leishmaniaasi koskettaa enimmäkseen hyvin köyhiä ihmisiä kehitysmaissa. Leishmaniaasia on neljä päätyyppiä, joista yksi, viskeraalinen leishmaniaasi, on hoitamattomana tappava. Viskeraalista leishmaniaasia aiheuttaa L. donovani Itä-Afrikassa ja Intian niemimaalla sekä L. infantum Välimeren ympäristössä ja eteläisessä Amerikassa. Leishmania-parasiitilla on kaksi muotoa: flagellallinen promastigoottimuoto, joka elää hietakärpäsen vatsassa, sekä flagellaton amastigoottimuoto, joka elää ja lisääntyy valkosolujen sisällä. Kaikilla nykyään käytössä olevilla lääkkeillä on vakavia haittavaikutuksia ja ne tulee antaa terveydenhoidon ammattilaisten valvonnassa. Myös resistenssiä esiintyy kasvavaan tahtiin. Uusille lääkkeille on siis todellinen tarve. Tämän työn tarkoituksena oli syntetisoida pieni molekyylikirjasto bentsoksadiatsolijohdannaisista, koska rakenteeltaan samankaltaisilla yhdisteillä oli aiemmin huomattu olevan aktiivisuutta L. donovania vastaan. Nämä yhdisteet oli tarkoitus testata yhteistyölaboratoriossa in vitro -kokeilla L. donovanin amastigoottimuotoa vastaan. Kirjastoa varten onnistuttiin syntetisoimaan 10 johdannaista, joista osa syntetisoitiin aiemmin raportoidun seitsenvaiheisen synteesireitin avulla, osalle suunniteltiin uusi synteesireitti. Kaikki synteesireitit ja suunnitelmat on kuvailtu tarkoin tässä työssä. Valitettavasti aktiivisuustestien tuloksia ei ehditty saada tämän työn kirjoittamiseen mennessä, joten rakenne-aktiivisuussuhteita ei voitu käsitellä. Työssä on kuitenkin pohdittu minkälaista tietoa syntetisoitujen yhdisteiden avulla voidaan mahdollisesti saada rakenteeltaan samankaltaisen yhdisteiden L. donovanin vastaisesta aktiivisuudesta. |
