Abstract (eng): | Bacterial biofilms deteriorate the operation of many industrial processes. Knowledge of the surface properties of microbial cells is a key to gain detailed understanding of their functions in natural environment and of the damages they cause in industrial processes. Deinococcus geothermalis is known to be a primary biofilm former in the paper machine water. It produces an adhesion platform for secondary bacteria. The slimes, which are a result of the formation of biofilms cause severe damages in the paper making process. In this thesis the working principle and structure of the Atomic Force Microscope (AFM) are described, microbiological applications of AFM are reviewed, a sample preparation strategy for force measurements on D. geothermalis bacteria is developed and force measurements are performed. The results of the force measurements indicate that the biofilm deforms as the cantilever with a particle attached to it is detached from the surface of the biofilm. The elastic and adhesion properties of a D. Geothermalis bacterium and the extracellular polymeric substances (EPS) are different. The results also suggest that the adhesion of D. geothermalis is quite strong, depending on the spot on the biofilm, where the force curve is generated. |
Abstract (fin): | Biofilmien kasvu haittaa monia teollisia prosesseja, kuten paperinvalmistusta. Mikrobien ja bakteerisolujen pintaominaisuuksien tunteminen on ehdottoman olennaista, jotta voitaisiin ymmärtää mikrobien toimintaa luonnossa sekä niiden aiheuttamia haittoja teollisissa prosesseissa. Deinococcus geothermalis on niin sanottu primaaritarttuja, joka muodostaa tartunta-alustan muille bakteereille. Biofilmin kasvun seurauksena muodostuvat limat aiheuttavat ongelmia paperinvalmistusprosessissa. Tässä diplomityössä selvitetään atomivoimamikroskoopin (AFM) rakenne ja toiminta sekä tarkastellaan atomivoimamikroskoopin soveltuvuutta mikrobiologisiin tarkoitusperiin kirjallisuuden pohjalta. Tämän työn aikana myös kehitettiin näytteenvalmistusmenetelmä, joka mahdollisti voimamittauksien suorittamisen D. geothermalis-bakteereilla ja vuorovaikutusvoimia mitattiin tämän bakteerin ja teräksen välillä. Tuloksista voidaan päätellä, että D. geothermalis biofilmi venyy kantileverin vetäytyessä pinnasta poispäin ja irrotessa siitä. Biofilmin elastiset ja tarttumisominaisuudet ovat erilaiset eri kohdissa biofilmiä. Tartuntavoiman suuruus riippuu siis ilmeisesti siitä, missä kohdassa biofilmiä mittaus suoritetaan. Yleisesti ottaen tartuntavoimat ovat kuitenkin suuria. |
