Abstract (eng): Propionic acid is a carboxylic acid used mainly for preservation of foods.
In industry, propionic acid is mainly produced by the hydrocarboxylation of ethylene.
Propionic acid is also the main metabolic product of propionic acid bacteria.

The literary review deals with metabolic routes of propionic acid bacteria, propionic acid inhibition and methods used to increase propionic acid concentration and volumetric productivity in fermentation.
Experimental part consists of experiments carried out at laboratory of Bioprocess Engineering at Helsinki University of Technology.
Propionibacterium acidipropionici NRRL B-3569 was used for fermentations.
The object of the experimental part was to create a bioprocess to produce propionic acid with high volumetric productivity and end product concentration.

P.acidipropionici cells were immobilized in a column filled with porous polypropylene cloth.
When media was fed continuously trough the column high volumetric productivities up to 7 g/l h were achieved, but propionic acid concentration remained low.
Problem was caused by a pH drop to 4,5.

Problem was solved by scaling up the column size to 900 ml and connecting the column to a fermentor.
Temperature and pH were controlled in the fermentor and media was recycled through the column.
Propionic acid concentration of 32 g/l from sucrose and 29.5 g/l from glycerol were achieved in a continuous process.
Volumetric productivities varied between 2-4 g/l h.
Closed loop experiment produced concentration of 78 g/l.
Abstract (fin): Propionihappo on kolmehiilinen monokarboksyylihappo.
Propionihappoa käytetään erityisesti ruokien ja rehun säilömiseen.
Propionihappoa valmistetaan pääasiassa petrokemiallisesti eteenistä.
Myös propionihappobakteerit tuottavat propionihappoa päämetaboliatuotteenaan.

Kirjallisuusosassa käydään läpi propionihappobakteereiden metaboliareittejä eri substraateista, ja erilaisia syitä, joista propionihapon aiheuttama inhibitio johtuu.
Lisäksi esitellään erilaisia menetelmiä, joilla propionihapon loppukonsentraatiota ja volumetristä tuottonopeutta on yritetty parantaa.
Tutkimusosassa käydään lävitse kokeita, jotka suoritettiin Teknillisen korkeakoulun bioprosessitekniikan laboratoriossa.
Propionibacterium acidipropionici NRRL B-3569 valittiin tuotantobakteeriksi.
Tutkimusten tavoitteena oli kehittää fermentaatioprosessi, jossa propionihapon volumetrinen tuottonopeus ja loppupitoisuus olisivat mahdollisimman suuria.

Soluimmobilisaatio huokoiseen polypropyleenikuitukankaaseen valittiin tuotantomenetelmäksi.
Immobilisaatio kantajaan pystyttiin todistamaan käyttäen nestetilavuudeltaan 50 ml kolonneja.
Nämä kolonnit antoivat myös korkeat volumetriset tuottonopeudet, jotka maksimissaan olivat yli 7 g/ l h, mutta tuotteen loppupitoisuus oli matala.
Ongelma johtui pH-arvon laskemisesta kolonnissa.

Suljetulla kierrolla kolonni saavutti 78 g/l propionihappopitoisuuden.
