Abstract (eng): Diabetic neuropathy is common for circa 50% of patients with type I or II diabetes.
Neuropathic pain causes allodynia, dysesthesia, hyperalgesia and hyperpathia.
The focus of the Master's thesis is on effects of diabetic neuropathy on synaptic area of primary afferent nociceptive neurons and spinal cord substantia gelatinosa neurons.

The hypothesis is that diabetic neuropathy increases the activity of substantia gelatinosa neurons of diabetic test animals compared to healthy test animals.
The literature part consists of theoretic background and the essential earlier research.

Setting up of the tissue slice patch clamp equipment is described in the experimental part.
The equipment is used to test substantia gelatinosa neurons.
Future research possibilities and effects of TRPA1 ion channel are discussed.
Possibility to use TRPA1 as a target protein for pharmacotherapy is also considered.
Abstract (fin): Diabeettista neuropatiaa havaitaan noin 50% tyypin I tai II diabetesta sairastavilla potilailla.
Neuropatia aiheuttaa hermoston herkistymistä ärsykkeille, allodyniaa, dysestesiaa, hyperalgesiaa, hyperpatiaa ja tuikekipua.
Diplomityössä keskitytään diabeettisen neuropatian vaikutuksiin selkäytimen substantia gelatinosan ja takajuuren kautta selkäytimeen tulevien primäärien afferenttien kipuhermojen synapsialueen aktiviteetissa.
Hypoteesina on diabeettisesta neuropatiasta kärsivien koe-eläinrottien substantia gelatinosan neuronien suurempi spontaani aktiivisuus verrattuna terveiden verrokkieläinten neuronien aktiivisuuteen.
Kirjallisuuskatsausosa esittää teoreettisen taustan kokeellisessa osassa rakennettavalta laitteistolta vaadittaville ominaisuuksille ja mittausten suorittamiselle sekä aiemman keskeisen aiheeseen liittyvän tutkimuksen.

Diplomityön kokeellisessa osassa pystytetään leike-patch-clamp-laitteisto substantia gelatinosan neuronien leikenäytteiden sähköfysiologisia mittauksia varten ja suoritetaan koesarja laitteiston ja tutkimushypoteesin testaamiseksi.
Lopuksi arvioidaan laitteiston soveltuvuus diabeettisen neuropatian vaikutusten tutkimukseen ja esitetään jatkotutkimusmalleja neuronien aktiivisuuteen vaikuttavien TRPA1-kanavien merkityksen tarkentamista varten huomioiden kanavan mahdollisuus kipulääkityksen kohdeproteiinina.
