Abstract (eng): | Calyculins, highly cytotoxic polyketides, originally isolated from marine sponge Discodermia calyx by Fusetani and co-workers, belong to the group of Lithistid sponges. These molecules have become an interesting target for cell biologists and synthetic organic chemists. The serine/threonine protein phosphatases play an essential role in the cellular signalling, metabolism, and cell cycle control. Calyculins express potent protein phosphatase 1 and 2A inhibitory activity, and have therefore become valuable tools for cellular biologists studying intracellular processes and their control by reversible phosphorylation. Calyculins might also play an important role in the development of several diseases such as cancer, neurodegenerative diseases, and type 2- diabetes mellitus. The fascinating structures of calyculins have inspired various groups of synthetic organic chemists to develop total syntheses of the most abundant calyculins A and C. However, with fifteen chiral centres, a cyano-capped tetraene unit, a phosphate-bearing spiroketal, an anti, anti, anti dipropionate segment, an alpha-chiral oxazole, and a trihydroxylated gamma-amino acid, calyculins reach versatility that only few natural products can surpass, and truly challenge chemists' knowledge of modern asymmetric synthesis. The literature review of this thesis discusses the role of protein phosphatases, naturally occurring toxins that inhibit protein phosphatases focusing on calyculins their origin and binding properties. The previous efforts, their advantages and downsides, towards total synthesis of calyculin A, B, and, C were also discussed in detail. The experimental part of this thesis focused on synthesis of C1-C8 tetraene subunit of calyculin C in detail on synthesis of C1-C5 diene. The key ideas of the synthesis were to construct vinyl nitrile with a good leaving group and further use Pd-catalyzed cross coupling to form the diene 33. |
Abstract (fin): | Kalikuliinit ovat ryhmä hyvin sytotoksisia polyketideitä ja niitä eristi ensikerran Fusetani et. al. meressä esiintyvästä sienestä Discodermia calyx, joka kuuluu Lithistide sieniin. Näistä molekyyleistä on tullut kiintoisa kohde solubiologeille ja synteettisille orgaanisille kemisteille. Seriini/treoniini proteiinifosfataaseilla on tärkeä rooli solujen signaalien välityksessä, metaboliassa ja solusyklin kontrolloinnissa. Kalikuliinien on todettu olevan potentiaalisia proteiini fosfataasiinhibiittoreita, ja siksi niistä on tullut tärkeä työkalu solubiologeille tutkittaessa solujen sisäisiä prosesseja ja niiden kontrollointia reversiibelillä foforylaatiolla. On myös mahdollista, että kalikuliineilla voitaisiin vaikuttaa useiden sairauksien, kuten syövän, neurologisten rappeumasairauksien sekä tyypin 2 diabeteksen synnyssä. Kalikuliinien, erityisesti keskeisimpien kalikuliini A:n ja C:n, kiehtova rakenne on innoittanut useita synteettisen orgaanisen kemian ryhmiä kehittämään kokonaissynteesiä näille molekyyleille. Näiden molekyylien rakenteessa on kuitenkin viisitoista kiraliakeskusta, syaani päätyinen tetraeeni ryhmä, fosfataasi-spiroketaali, anti, anti, anti dipropionaatti osa, 0-kiraalinen oksatsoli ja trihydroksyloitu 0-aminohappo. Harvan luonnonaineen rakenne yltää monimutkaisuudessaan tälle tasolle. Tämä rakenne todella haastaa kemistin modernin asymmetrisen synteesin tietämyksen. Tämän työn kirjallisessa osassa tarkastellaan proteiinifosfataasien roolia, sekä niitä inhiboivien luonnossa esiintyvien toksiinien, erityisesti kalikuliinien, alkuperää, ja sitoutumisominaisuuksia. Lisäksi tarkastellaan kalikuliini A, B ja C:n aikaisempia kokonaissynteesejä, niiden vahvuuksia ja heikkouksia. Työn kokeellisessa osassa keskityttiin tetraeeni C1-C8:n ja erityisesti C1-C5 dieenin synteesiin. Keskeisenä ajatuksena oli syntetisoida vinyyli nitriili molekyyli, jossa olisi myös hyvä lähtevä ryhmä, jolloin voitaisiin käyttää Pd-katalysoitua kytkentää dieeni 33:n syntetisointiin. |
