Abstract (eng): | Compared to e.g. oral medication, transdermal drug release has various benefits. As the drug is administered through the skin, it avoids enzymatic degradation in the digestive tract and the first pass metabolism. Additionally, transdermal drug release allows more constant and controlled drug administration. Since particularly the Stratum corneum, the outermost layer of the skin, is a formidable hydrophobic barrier, not all drugs can be administered transdermally. One of the developed enhancement methods is iontophoresis, in which drug transportation is enhanced by a low, painless electric current. Iontophoresis enhances drug release by migration, but it is also based on electro-osmosis (flow of fluid which is independent of concentration gradient) and changes in skin permeability. This study continues the research of a novel iontophoretic half-cell prototype developed at the Department of Chemistry. It uses Smopex®-102 ion exchange fiber into which the drug has been loaded. Ion exchange fibers contain charged functional groups, which enable exchanging preloaded counter ions with ions surrounding the fiber in an external phase complying with the electroneutrality principle. The effect of NaCl and CaCl2 solutions of different concentrations in the drug department on drug release rate was studied. The drugs used in the study were metoprolol and tacrine. The experiment results suggest that CaCl2 is not a good candidate for electrolyte solution per se because especially at larger CaCl2 concentrations iontophoretic enhancement was non-existent. Also constant flow was not observed at the larger concentrations; the cumulative flux complied with Higuchi equation. However, iontophoretic enhancement at lower concentrations was observed. |
Abstract (fin): | Ensikierron metabolia ja ruoansulatuskanavan entsyymit heikentävät useiden oraalisesti nautittavien lääkkeiden lääkinnällistä vaikutusta. Nämä ongelmat vältetään, kun lääke annostellaan ihon läpi. Transdermaalinen lääkintä mahdollistaa myös tasaisemman ja kontrolloidumman lääkeainekuljetuksen, jolloin lääkeainekonsentraatio voidaan helpommin säilyttää terapeuttisessa ikkunassa. Ihon stratum corneum (marraskesi) on vaikeasti läpäistävä hydrofobinen este, joten kaikkia lääkeaineita ei voida kuljettaa passiivisesti ihon läpi. Yksi kehitetyistä tehostusmenetelmistä on iontoforeesi, jossa lääkeaineen kulkeutumista tehostetaan matalalla, kivuttomalla sähkövirralla. Menetelmä tehostaa varattujen lääkeaineen kulkeutumista migraatiolla, mutta se perustuu myös elektro-osmoosiin (sähkövirran aiheuttama konsentraatiogradientista riippumaton nestevirtaus) sekä ihon permeabiliteetin muutoksiin. Tässä työssä jatkettiin Kemian laitoksella kehitetyn iontoforeettisen puolikennoprototyypin optimointia. Kenno hyödyntää Smopex®-102-ioninvaihtokuitua, johon lääkeaine on ladattu. Ioninvaihtokuidut ovat varauksellisia funktionaalisia ryhmiä sisältäviä kuituja, jotka pystyvät kiinteä-neste-faasireaktion avulla sitomaan ja vapauttamaan varattuja osaslajeja elektroneutraalisuusperiaatteen mukaisesti. Työssä tutkittiin, miten eri konsentraatioiset NaCl- ja CaCl2-liuokset puolikennon lääkeainetilassa vaikuttavat kuljetuksen onnistumiseen. Iontoforeesikokeita tehtiin takriini- ja metoprolol-lääkeaineilla. CaCl2 ei sellaisenaan osoittautunut kelpaavaksi, sillä, iontoforeettinen tehostus jäi varsinkin suurilla konsentraatioilla olemattomaksi; NaCl:ia käytettäessä iontoforeettinen tehostus oli selkeästi havaittavissa. Suuret elektrolyyttikonsentraatiot saattoivat myös aiheuttaa Higuchi-riippuvuutta, mikä ei ole toivottavaa transdermaalisessa lääkeannostelussa. |
