Abstract (eng): | Enteroviruses are RNA viruses known to cause several diseases e.g. hand, foot and mouth disease or aseptic meningitis but usually the infection is assymptomatic and self limitting. Enteroviruses are also associated as one causative agent in the initiation of type 1 diabetes (T1D), an autoimmune caused functional loss of insulin producing beta-cells in human pancreas. The Babydiab is a study which tries to reveal the possible association of enterovirus infections and early introduction of gluten to the diabetes development. In this master's thesis several stool samples are analyzed for enteroviruses from children with higher genetic risk of developing T1D. Samples where analyzed with several PCR based methods and virus propaganition in RD(A) cells. Enterovirus real-time-PCR positive detection rate was 9.6 % and in 94 % of these samples the enterovirus matching sequence data could he identified. Total of 32 enteroviruses where identified in 9.3 % of samples from 23 children. The four most prominant viruses found where CAV-4, CAV-2, CAV-10 and echo-25. The seasonal variation of enterovirus positive samples was same as in the literature, positive samples peaking at July and September. The Turku pilot is a study which tries to examine the suitability of current real-time-PCR method for detecting upper respitary track infection associated viruses from nasal swap samples. 1 detected human enteroviruses with real-time-PCR in 5.5 % of nasal swap samples. All the positive samples where taken from people suffering from some kind of infection so that 15 % of these sick people where enterovirus positive in real-time-PCR. Based on these results several different enteroviruses serotypes are common source of different infections. |
Abstract (fin): | Enterovirukset ovat RNA viruksia, jotka voivat aiheuttaa useita tauteja, kuten aseptista aivokalvontulehdusta tai enterorokkoa. Yleensä enterovirusinfektio on kuitenkin oireeton ja itsestään parantuva. Enterovirukset on myös yhdistetty yhdeksi tekijäksi tyypin 1 diabeteksen (T1D) puhkeamisessa. Babydiab on tutkimus jossa pyritään selvittämään geneettisesti korkeassa diabetesriskissä olevien lasten enterovirusinfektioiden ja ensimmäisen gluteenialtistuksen assosiaatiota diabeteksen kehitykseen. Diplomityössäni käytiin läpi näiden lasten ulostenäytteitä enterovirusten detektoimiseksi. Tutkimuksessa osoitettiin, että 9,6 % näytteistä enteroviruspositiivisiksi real-time-PCR:llä. Näistä näytteistä 94 % pystyttiin tyypittämään serotyyppitasolle siten, että yhteensä 32 enterovirusta löydettiin 9,3 % näytteistä 23 lapselta. Neljä eniten löydettyä serotyyppiä olivat CAV-4, CAV-2, CAV-10 ja echo-25. Positiiviset näytteet noudattivat enteroviruksille tyypillistä vuodenaikaisjakaumaa siten, että eniten positiivisia näytteitä löydettiin heinä- syyskuussa. Tutkimuksen tulokset ovat yhteneväisiä kirjallisuuden kanssa ja tulosten perusteella useat enterovirukset voivat aiheuttaa infektioita lapsilla Diplomityöni on myös osa Turun pilotti -tutkimusta, jonka yksi tarkoitus oli selvittää olemassa olevan realtime-PCR:n soveltumista ylähengitystieinfektioita aiheuttavien virusten detektoimiseksi nenänielunäytteistä. Diplomityössäni osoitin 5,5 % näytteistä enteroviruspositiivisiksi real-time-PCR:llä siten, että kaikki positiiviset näytteet olivat sairailta henkilöiltä. Täten sairaista henkilöistä 15 % oli enteroviruspositiivisia. Tulosten perusteella ylähengitystieinfektioihin liittyvät enterovirukset voidaan osoittaa nenänielunäytteistä real-time-PCR:llä. |
